Preclinical Xenograft and Oncology Studies

Perry Scientific’s preclinical oncology group has more experience with oncology and xenograft studies than virtually any other CRO. We create literally thousands of xenograft models each year in nude mice, SCID mice and nude rats. Perry Scientific is also a leader in the evaluation of new cancer agents in mice and rats exhibiting chemotherapy or radiation induced neutropenia.

Perry Scientific’s scientists have extensive experience with the design and execution of efficacy studies for all types of experimental cancer therapeutic agents including both small molecules and biologics. Our expertise includes evaluation of anti-neoplastic, anti-angiogenesis agents, monoclonal antibodies, immune system stimulators and other experimental agents.

Perry Scientific can follow your custom protocol, design one for you after consultation with your scientists, or use standard NCI protocols.

 

Tumor Models

Perry Scientific can produce both human xenografts and syngenic tumors in rats and mice. Xenografts can be grown in nude mice, SCID mice or nude rats and we can evaluate primary tumor growth or metastasis. Syngenic tumors can be grown in most species of mice. We offer traditional subcutaneous models as well as orthotopic and other models and can customize models to meet a sponsor’s needs. We have experience with most of the NCI human tumor lines. We carry many of them in our own liquid nitrogen storage, and can obtain others at short notice.

We routinely create and evaluate the following animal models:

  • Mammary fat pad models
  • Custom model development

We use the following rodents:

  • Nude mice (human xenografts)
  • SCID mice (human xenografts)
  • Normal mice (syngenic tumors)
  • Immunodeficient rats (human xenografts)
  • Normal rats (syngenic tumors)

We use the following study end points:

  • Primary tumor growth
  • Metastasis
  • Metastatic growth
  • Mortality
  • Metastasis evaluated by MRI scanning

We can utilize any dose schedule or dosing route including:

  • Subcutaneous
  • Intraperitoneal
  • Intravenous
  • Oral

 

Tumor Lines

Human Tumor Xenograft cell lines commonly used at Perry Scientific include:

  • Breast: MDA MB-231, MCF7, MDA-MB-435, T-47D
  • Colon: KM12, HCT-15, COLO 205, HT29
  • Lung: NCI-H460, A549
  • Prostate: CRW22, LNCAP, PCC-3, DU-145
  • Melanoma: LOX-IMVI
  • Ovarian: SK-OV-3
  • Renal: CAKI-1, A498, SN12C

In addition to these, we can form almost any xenograft you require. Perry Scientific’s library of human tumor and syngenic cell lines are available for all common malignancies. Other cell lines, which are commercially available or supplied by the sponsor, can also be used, following evaluation for growth characteristics.

 

Perry Scientific is a leader in the evaluation of new cancer agents using neutropenia models. We can produce either mouse or rat modes of:

  • Chemotherapy induced neutropenia
  • Radiation induced neutropenia
  • Combination radiation and chemotherapy induced neutropenia

Study Design

The receipt of a completed protocol from the customer may start the project. Alternatively, Perry Scientific can meet with your scientists, either in person or over the telephone, to discuss the requirements for the study. Protocol details are then established, including methods of drug preparation and delivery.

 

Pathology

Perry Scientific performs necropsies as well as almost any type of pathology. We have facilities to fix, cut and stain any number of slides for histology. These prepared and stained slides can be returned to the customer for evaluation, or they can be read by our consulting animal pathologist.
